Disturbance of hepatic lipid metabolism, insulin resistance, oxidative stress, or inflammation can contribute to hepatic steatosis, liver injury, fibrosis, and metabolic dysfunction–associated steatotic liver disease (MASLD).
The liver is also an important therapeutic target. Drug-induced cytotoxicity and organ toxicity are assessed in relation to hepatic function, while emerging genetic medicines may use lipid nanoparticles or other delivery systems to modify hepatocyte genes. The organ has a central role in transplantation and abdominal surgery because of its vascular anatomy, size, and capacity to support whole-body metabolism.
