CD44/JAK2/STAT3 signaling pathway

The CD44/JAK2/STAT3 signaling pathway is a cell-Signaling axis centered on the transmembrane adhesion receptor CD44 and the intracellular kinases/transcription factors JAK2 and signal transducer and activator of transcription 3 (STAT3).

Where the papers sit

11 papers from the last 6 months study CD44/JAK2/STAT3 signaling pathway directly · updated from PubMed 12 Oct 2026

Those 11 do not share a common subject.

  • Cancer therapies, inflammatory disorders, infection-related immunosuppression, and drug-safety assessment appear without a shared mechanistic question. Cancer studies test targeted combinations, immunotherapy, nanoparticles, and EGFR/JAK2/STAT3 inhibition, while inflammatory studies focus on STAT3/NF-κB or Th17 modulation. Evidence ranges from cell lines and animals to a QT study, so no consistent therapeutic direction emerges. The cluster is best treated as a heterogeneous collection rather than a developing research program.

PARADIGM SHIFT

Durable treatment cannot rely on STAT3 blockade as an isolated therapeutic intervention

The fungal-metabolite immunotherapy study in tumor-associated macrophages and the multi-node targeted therapy study in orthotopic pancreatic ductal adenocarcinoma independently point beyond single-axis STAT3 inhibition: IM502 overcomes immunosuppression and PD-1-blockade resistance by inhibiting PI3Kγ and redirecting signaling from STAT3/6 toward STAT1/2, whereas combined KRAS, EGFR-family, and STAT3 inhibition produces complete tumor regression without resistance 42173096May 42224594Jun. The resulting change is to treat STAT3-associated resistance as a network or state problem requiring signaling reprogramming or simultaneous blockade of parallel dependencies, rather than as a defect that can reliably be solved by suppressing STAT3 alone.

NEW DIRECTION

CD44 can act as a glycan-dependent antibody decoy that physically limits target access

The chondroitinase ABC study in pancreatic ductal adenocarcinoma assigns CD44 a role not represented by the set’s CD44-targeted delivery systems: CD44 was identified as a core protein of chondroitin sulfate A-bearing proteoglycans that mask HER2, while the CS-A chains also bind trastuzumab and inhibit its engagement with HER2 42001719Apr. This extends CD44 from a targeting receptor or signaling-associated molecule to a structural mediator of antibody resistance, suggesting that enzymatic removal of CD44-associated glycans may restore antibody access where drug loading or receptor-directed delivery alone does not.
