Anti-inflammatory cytokines
Anti-inflammatory cytokines are immunoregulatory proteins that dampen, resolve or counterbalance responses driven by pro-inflammatory mediators such as interleukin-1 beta (IL-1β), Interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α).
Anti-inflammatory cytokines are immunoregulatory proteins that dampen, resolve or counterbalance responses driven by pro-inflammatory mediators such as interleukin-1 beta (IL-1β), Interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α). Interleukin-10 (IL-10) is the archetype, alongside transforming growth factor-beta (TGF-β), interleukin-4, interleukin-13 and interleukin-35, together with the receptor antagonist IL-1Ra, which suppresses by occupying a receptor rather than by signaling through one. They act through receptor-mediated cascades — including the JAK2/STAT3 signaling pathway and PI3K/Akt signaling pathway — to restrain immune cell activation, direct macrophage polarization toward reparative states, and promote tissue repair. IL-10 in particular signals through STAT3 to shut down transcription of inflammatory genes in myeloid cells, and its necessity is demonstrated by loss-of-function mutations in IL-10 or its receptor, which cause severe infant-onset inflammatory bowel disease.
The labels are contextual rather than intrinsic. TGF-β suppresses immune activation while also driving the fibrosis that destroys tissue in chronic disease, IL-4 and IL-13 restrain type 1 inflammation while driving allergy, and several of these cytokines promote tumor growth by suppressing the antitumor response — so the same molecule can be protective or pathogenic depending on tissue and timing.
The balance between the two arms shapes outcome across autoimmune, metabolic, neurodegenerative and malignant disease, and deficient anti-inflammatory cytokine signaling is associated with inflammatory bowel disease, rheumatoid arthritis, psoriatic arthritis and cardiovascular disease. Restoring or amplifying that signaling is a longstanding therapeutic aim, but a difficult one in practice: systemic recombinant IL-10 largely failed in Crohn's disease despite a compelling rationale, limited by a short half-life and by adverse effects associated with systemic administration, despite its ability to act throughout the circulation. Current approaches accordingly favor local or cell-based delivery, pharmacological induction, and engineered variants with altered receptor selectivity.

Inflammatory Cytokine Profiling
Inflammatory cytokine profiling links CXCL8, IL-6 and broader mediator panels to disease state across cancer progression, chemotherapy, and severe traumatic brain injury. The work measures systemic and compartment-specific responses, including blood and cerebrospinal fluid, rather than converging on one intervention. Exercise during chemotherapy is evaluated for effects on serum cytokines and muscle autophagy, while obesity-related expression is examined in cervical cancer. Dynamic timing and context remain central, with no shared treatment direction or consistent cytokine signature established.

Targeted RNA and Tissue Repair
Targeted RNA delivery and biomaterial repair are moving toward localized control of inflammation and tissue damage. Lipid nanoparticles are being tuned for lung delivery and nose-to-brain mRNA transport that bypasses the blood-brain barrier, with BDNF and cognitive function as brain-repair readouts. A cardiac microneedle patch instead addresses post-infarction immunity and fibrosis through spatially timed intervention. The shared direction is safer, more precise delivery, but the platforms and disease endpoints remain distinct.

Others — Inflammation in Disease and Repair
Immune regulation appears in chronic wound healing, pain-related TMD, and antitumor therapy, but these contexts do not form one translational program. Biomimetic or antibacterial hydrogels combine matrix remodeling, anti-inflammatory activity, and pro-angiogenic support for diabetic wounds. Twin comparisons examine inflammatory, oxidative, and neurotrophic profiles, while anti-VEGFR-2/PD-1 therapy links IL-10 secretion by CD8 T cells to early NK recruitment. The cluster therefore has no single direction beyond context-specific modulation of inflammation and repair.

IL-10 can promote antitumor immunity by coordinating immune-cell recruitment
The anti-VEGFR-2/PD-1 therapy study in tumors found that IL-10 secreted by CD8 T cells orchestrates early NK-cell recruitment and antitumor immunity, rather than serving only as a suppressive or reparative mediator of inflammation 42331382Jun. This assigns an active immune-organizing role to an anti-inflammatory cytokine: IL-10 becomes part of the mechanism that mobilizes effective antitumor responses after combined antiangiogenic and checkpoint therapy.
