• PD-L1 Immunotherapy Strategies

  PD-1/PD-L1 blockade is expanding beyond conventional antibodies toward small molecules, masked conjugates, STING agonists, microbiome interventions, and pyroptosis-based immune activation. Colorectal cancer, glioblastoma, and other tumors recur as settings for altering PD-L1 trafficking, regulation, or immune sensitivity. Microbial composition, miRNA patterns, genomic features, and respiratory microbiota are being tested as response or toxicity biomarkers. Most approaches remain preclinical, leaving clinical validation and patient selection open.

• Lung Cancer Biomarker Assessment

  PD-L1 evaluation in non-small-cell lung cancer is moving toward automated, multi-scale measurement alongside treatment selection. H&E annotations and chest CT models use masked autoencoders and GAN augmentation to predict PD-L1 expression. Gilteritinib is separately being studied to overcome resistance to second-generation TKIs in ALK-rearranged disease. Hypoxia, TP53, and glycolytic features frame the tumor biology, but the cluster has not established a single clinical endpoint.
