AKT1
AKT1, also known as protein kinase B (PKB), is a serine–threonine protein kinase that functions within the PI3K/AKT/mTOR signaling pathway.
AKT1, also known as protein kinase B (PKB), is a serine–threonine protein kinase that functions within the PI3K/AKT/mTOR signaling pathway. It is associated with regulation of tumor-cell proliferation, survival, metabolism, and therapeutic resistance, making it a frequently investigated molecular target in cancer biology. Recent studies have examined AKT1 in pan-cancer analyses, head and neck cancer risk prioritization, colorectal and breast cancer cytotoxicity models, non-small-cell lung cancer, and intrahepatic cholangiocarcinoma. These investigations have used approaches including molecular docking, proteomics, genetically informed analysis, and evaluation of integrative therapies.
AKT1 is also studied in cardiovascular and inflammatory contexts. Research has examined its phosphorylation at serine 473 in relation to finerenone-mediated effects in diabetic cardiomyopathy, and has assessed AKT1 together with eNOS, STAT3, GPX4, and other components of oxidative-stress and ferroptosis-related pathways in myocardial injury and heart failure. Other work has investigated AKT1 within an AKT1–STAT3–TLR4 axis linked to autophagy in atherosclerotic comorbid depression. Collectively, these studies place AKT1 at the intersection of signaling research involving PI3K, mTOR, STAT3, KRAS, autophagy, oxidative stress, ferroptosis, and inflammatory or vascular injury, while reflecting diverse experimental and therapeutic contexts rather than a single established treatment strategy.

AKT1 or PI3K-AKT signaling recurs across inflammation, cancer, cardiovascular disease and neurological injury. The interventions range from plant-derived compounds and traditional formulations to finerenone and computational drug design. Endpoints include survival, inflammation, autophagy, vascular injury and tumour growth. The diseases and therapeutic questions are too disparate to support a common direction.
AKT1 is elevated to a genetically supported immune-checkpoint susceptibility factor in head and neck cancer
The head and neck cancer study using summary-data Mendelian randomization and multi-omics QTLs found AKT1 to be a risk-associated, genetically prioritized immune-checkpoint gene, with consistent effects in sensitivity analyses and no evidence of horizontal pleiotropy. Whereas the other studies use AKT1 mainly as a signaling mediator, tumor biomarker, prognostic marker, or drug target, this finding assigns it a distinct role as an inherited susceptibility factor linked to immune-checkpoint biology, shifting AKT1 prioritization toward genetic risk assessment in head and neck cancer 42773293Sep.
