  • Cellular Senescence and Tissue Aging

Senescence is being treated as a modifiable driver of tissue decline, impaired regeneration and treatment response. IGFBP5 and non-canonical Wnt signaling reverse dental follicle stem-cell senescence, while hepassocin activates AMPK to support liver regeneration. LSD1-mediated ATM demethylation links DNA-damage signaling to organ aging, and therapy-induced senescence in acute myeloid leukemia may increase immunogenicity through reduced EZH2 activity. The work is moving toward pathway-specific strategies that either prevent senescence or exploit it therapeutically.

  • Inflammation and Tissue Aging

Inflammatory signaling is being targeted alongside tissue repair and cellular aging. Biomimetic and platelet-membrane vesicles deliver resveratrol or other payloads to limit neuroinflammation, mitochondrial dysfunction and endothelial senescence. Macrophage polarization and redox-inflammatory signaling recur as links between local injury and broader tissue decline. Glycine studies add a systemic perspective, associating slower biological aging with sex- and diet-dependent inflammatory effects, but their evidence is less interventional.

  • Senescence-Targeted Cancer Immunotherapy

Cancer research is moving toward using senescence as both a therapeutic target and a biomarker for immune treatment selection. Nanotherapies aim to eliminate or modulate senescent cells, while pan-cancer multi-omics seeks signatures that predict immunotherapy response. Human cytotoxic T-cell activity and the interaction between senescent cancer cells and immune surveillance remain central to translation. Clinical utility will depend on validating these signatures and showing that senescence-directed treatment improves survival rather than merely changing immune profiles.

PARADIGM SHIFT

Senescence can function as an antitumor immune state rather than merely a pathological state to eliminate

Acute myeloid leukemia subjected to chemotherapy and pan-cancer tumors analyzed by multi-omics both indicate that senescence can be associated with, and potentially strengthen, antitumor immunity rather than simply impair tissue function or promote disease. In AML, therapy-induced senescence increased interferon signaling, HLA expression, antigen presentation, T-cell activation, and sensitivity to checkpoint blockade; across 31 cancers, higher senescence scores were associated with greater CD8⁺ T-cell and NK-cell infiltration, immune cytolytic activity, and features relevant to immunotherapy response 42697881Sep 42722723Sep. This replaces the assumption that senescence is uniformly a detrimental state to suppress with a context-dependent view in which inducing or preserving senescence may sometimes improve immune recognition and cancer treatment.
