CTNNA1
CTNNA1 encodes catenin alpha 1 (α-catenin), a protein associated with catenin signaling.
CTNNA1 encodes catenin alpha 1 (α-catenin), a protein associated with catenin signaling. Recent research has examined CTNNA1 in the context of a germline mutation occurring with synchronous follicular and papillary thyroid carcinomas and numerous adenomatoid nodules; however, the relationship between CTNNA1 mutation and thyroid neoplasia remains poorly understood.
The broader literature places CTNNA1 within a disease-relevant catenin-signaling context that includes the Wnt/β-catenin pathway. Studies of related β-catenin signaling have investigated effects on tumor biology, including METTL14/Wnt9a-mediated cancer stem-cell activity, TCF/LEF-dependent transcription, and potential combinations with radiation therapy, immunotherapy, or anti-PD-1 therapy. Other work has linked modulation of this pathway to rheumatoid arthritis-associated bone erosion, diabetic osteoporosis and osteoblast ferroptosis, intestinal inflammatory signaling involving Toll-like receptor 4 and NF-κB, and immune-cell function in glioblastoma. These findings support continued investigation of CTNNA1 and catenin-network alterations in cancer, inflammation, and skeletal disease, while not establishing CTNNA1 itself as a validated therapeutic target.

β-Catenin Signaling in Disease
β-Catenin is emerging as a shared therapeutic hub across cancer, inflammation and bone loss. Studies target Wnt3a/Wnt9a–β-catenin signaling or alter β-catenin stability to improve radiotherapy, immunotherapy and protection from ferroptosis. Interactome mapping and mutation studies are expanding its mechanistic links to oncogenesis and tissue injury. The main unresolved issue is how to modulate β-catenin selectively across these contrasting disease contexts.
