  • MEK Inhibition in Cancer

MEK inhibition is moving toward treatment of resistant disease and more individualized testing. Nanoparticle drug combinations aim to improve efficacy in drug-resistant melanoma, while trametinib is assessed in glioblastoma avatars and pediatric brain tumors. Organoids and skin-on-a-chip models recur as tools for testing resistance and personalizing therapy. Uveal melanoma models suggest that clinically relevant resistance may be underestimated by standard cell lines.

  • RAS-Mutant Solid Tumors

Treatment is moving from trametinib alone toward molecularly selected combinations for RAS-mutant tumors. Clinical evaluation tracks partial response and stable disease in NRAS-mutated cancers, while SOS1 inhibition is being paired with trametinib in KRAS-positive solid tumors. PCDH7-targeting antibodies offer a separate strategy in KRAS-mutant non-small cell lung cancer by combining tumor inhibition with immune activation. The main open question is whether these combinations can improve the limited responses seen with pathway inhibition alone.

  • Others — Cancer Drug Resistance and Monitoring

Resistance is linked to MAPK-pathway alterations in anaplastic thyroid cancer and clonal evolution after quizartinib in leukemia. Another study focuses on measuring serum drug levels, shifting attention toward exposure and treatment safety. MAP2K1 and MEK1-related resistance recur, but the diseases and drugs do not form a single therapeutic program. The cluster therefore has no unified direction beyond connecting resistance biology with drug-response assessment.
