Glucosidase
Glucosidase refers to a group of carbohydrate-active enzymes that hydrolyze glycosidic bonds in glucosides and oligosaccharides, producing glucose or other sugar residues.
Glucosidase refers to a group of carbohydrate-active enzymes that hydrolyze glycosidic bonds in glucosides and oligosaccharides, producing glucose or other sugar residues. The term includes enzymes with different substrate specificities and stereochemical preferences, particularly α-glucosidases and β-glucosidases, which act on α- or β-linked glucose-containing substrates, respectively. Glucosidases are found in animals, plants, fungi, and microorganisms and participate in carbohydrate digestion, intracellular glycan processing, biomass degradation, and carbohydrate metabolism.
In human physiology, intestinal α-glucosidase activity contributes to the final stages of starch and disaccharide digestion by releasing glucose for absorption. Pharmacological inhibition of α-glucosidase can delay carbohydrate breakdown and glucose absorption, thereby reducing postprandial blood-glucose excursions. This mechanism has made α-glucosidase an established target in research on type 2 diabetes, often alongside amylase, which catalyzes earlier stages of starch hydrolysis. Acarbose is a commonly used reference inhibitor in biochemical and drug-discovery studies, while metformin has also been investigated for possible competitive interactions with α-glucosidase. Inhibitor discovery commonly uses enzyme-kinetic analysis, IC50 measurements, molecular docking, and assessment of hydrophobic interactions and hydrogen bonding.
Beyond diabetes research, glucosidases are being explored as targets for carbohydrate-processing technologies and enzyme-activated therapeutics. Selective enzymatic activation has, for example, been investigated as a means of triggering controlled radical release, while simultaneous inhibition of α- and β-glucosidase and amylase has been evaluated in experimental anticancer strategies.

Carbohydrate Enzyme Inhibition
α-Amylase and α-glucosidase inhibition is being pursued for postprandial glucose control and broader metabolic disease treatment. Work combines natural polysaccharides and plant metabolites with designed heterocycles, thiazolidinediones, benzothiazole conjugates, and carbohydrate-linked compounds. Dual or multitarget activity, supported by docking, DFT, ADME prediction, and biochemical assays, recurs across the studies. Metformin’s competitive inhibition and possible effects on carbohydrate metabolism add mechanistic context, but clinical translation and in vivo validation remain limited.
