  • CGRP Signaling in Pain

CGRP-targeted prevention is moving beyond efficacy trials toward long-term safety assessment in migraine patients with epilepsy or prior cerebrovascular and cardiovascular disease. Mechanistic work repeatedly connects CGRP to TRPV1, NaV1.7, neuroinflammation, vascular signaling, and receptor-degradation fragments. Botulinum toxin, botanical formulations, lavender, and bergenin are being explored as indirect ways to suppress this pain network. Evidence remains preliminary for non-migraine pain and for translating these pathways into broader therapies.

NEW DIRECTION

CGRP is used as a sustained regenerative signal rather than primarily as a migraine or pain mediator

The femoral condyle defect study using IL-4/CGRP-loaded GelMA microspheres assumed a therapeutic role for CGRP in a controlled-release biomaterial and found that sustained CGRP release enhanced angiogenesis and osteogenic differentiation, while coordinating with immune regulation to accelerate bone formation and defect healing. This shifts CGRP into an immuno-osteogenic and tissue-regenerative role distinct from the migraine and pain mechanisms used elsewhere in the set 42093387May.

NEW DIRECTION

CGRP degradation products are treated as functional local signals that can alter vascular, immune, receptor, and biomarker effects

The dura mater αCGRP degradation-fragment study assumed that local CGRP availability and signaling were insufficiently explained by intact peptide levels and found that degradation fragments themselves have functional relevance at vascular, immune, and CGRP-receptor targets, with implications for biomarker detection. This gives CGRP biology a distinct role in which peptide metabolism generates biologically consequential signals rather than merely terminating CGRP action 42168858May.
