Prostate cancer
Prostate cancer is a malignant disease of the prostate in which androgen receptor (AR) biology is an important focus of investigation, particularly in treatment-resistant and metastatic castration-resistant prostate cancer.
Prostate cancer is a malignant disease of the prostate in which androgen receptor (AR) biology is an important focus of investigation, particularly in treatment-resistant and metastatic castration-resistant prostate cancer. Current research examines AR expression, androgen-directed therapies such as enzalutamide and androgen deprivation therapy, and experimental AR-targeting heterobifunctional degraders. Proteomic studies have associated some phthalimide degraders with hepatotoxicity, highlighting the need to distinguish antitumor activity from treatment-related toxicity. Other molecular work is evaluating SAT1-mediated spermine catabolism as a factor in disease progression and reduced enzalutamide sensitivity.
Clinical management and assessment include prostate-specific antigen measurement, Gleason Score-based grading, active surveillance, radical prostatectomy—including robot-assisted radical prostatectomy—and radiotherapy. Recent studies address grade progression in relation to prostate volume and 5-alpha reductase inhibitor use, postoperative urinary incontinence, automated radiotherapy planning, and carbon-ion radiotherapy after rectal surgery. Emerging diagnostic and analytical approaches include seminal-fluid cell-free DNA methylation analysis for clinically significant disease and interpretable artificial intelligence models for distinguishing lower- from higher-grade tumors.

Prostate Cancer Risk and Care
Clinical decision-making is moving toward more individualized grading, progression risk, treatment selection, and survivorship outcomes. Interpretable multicenter machine learning and prostate volume or 5-alpha-reductase inhibitor data address grading and treatment decisions, while SAT1-mediated spermine catabolism is linked to enzalutamide resistance. Studies also examine postoperative urinary incontinence, radiotherapy-plan safety, and microbial genomes in patient samples. These approaches share a prostate-cancer focus but do not establish one unified biological direction.

Prostate Cancer Diagnostics and Treatment
Sulfonamide compounds are being evaluated for anticancer activity, while seminal-fluid cfDNA methylation is explored for detecting clinically significant disease. Carbon-ion radiotherapy is being assessed mainly for safety after rectal surgery. The shared focus is prostate cancer, but these studies do not converge on one mechanism, biomarker, or treatment strategy. Diagnostic validation and comparative clinical benefit remain open.

Androgen Receptor Targeting
Androgen receptor expression remains a central rationale for targeting prostate tumors. Heterobifunctional and phthalimide degraders are being developed to eliminate target proteins rather than merely inhibit them. Proteome-guided profiling is shifting attention toward hepatotoxicity, electron transport chain effects, and other off-target liabilities. Whether degradation can improve efficacy without unacceptable systemic toxicity remains open.

Prostate cancer is treated as a host-associated microbial ecosystem rather than solely as a tumor to diagnose, grade, treat, or monitor
The oral, blood, and prostate-tissue microbiome of patients with prostate cancer is the subject of the study, which reconstructs 249 highly complete bacterial genomes from whole-genome sequencing data and identifies previously unresolved oral bacterial species 42901952Oct. Whereas the other papers use prostate cancer as a disease whose tumor biology, treatment, toxicity, diagnosis, prognosis, or complications are being evaluated, this work makes the cancer setting a source for investigating microbial diversity and possible bacterial contributions to carcinogenesis. The resulting direction is metagenomic and causal investigation of host-associated microbes, not another application of the established tumor-centered frame.
