PIEZO1
PIEZO1 is a mechanosensitive ion channel protein that converts changes in membrane tension and mechanical forces into cellular signals, in part by promoting extracellular calcium ion influx.
PIEZO1 is a mechanosensitive ion channel protein that converts changes in membrane tension and mechanical forces into cellular signals, in part by promoting extracellular calcium ion influx. This mechanotransduction pathway can regulate calcium-dependent processes including Calpain-1 activity, cytoskeletal and focal adhesion signaling, cell aggregation, collective migration, and tissue remodeling. Recent structural work indicates that the PIEZO1 wedge domain contributes to state-dependent channel inactivation, including stabilization of a flat inactivated conformation, with wedge dissociation proposed to participate in recovery from inactivation.
PIEZO1-mediated signaling has been investigated in vascular, immune, wound-healing, and cancer contexts. Its activation has been associated with impaired brain microvascular endothelial cell tight junctions in high-altitude cerebral edema, mechanosensitive responses of macrophages linked to B-cell activation and IgM release, and stiffness-dependent scar formation. In tumor-related studies, the Caveolin-1/PIEZO1/PLA2G4A axis has been targeted to alter lipid metabolism in pancreatic cancer, while cholesterol-dependent PIEZO1 activation has been implicated in melanoma cell migration. Other work has examined PIEZO1 in the YAP1/angiopoietin-1 pathway during intraplaque neovessel maturation and used hydrogels or nanoparticles to modulate mechanically regulated calcium signaling.

Mechanosensitive Channel Biology
Mechanical and lipid cues are being connected to PIEZO1-dependent calcium entry, channel inactivation, and downstream changes in barrier integrity, migration, and inflammation. The work spans endothelial tight junctions and neovessel maturation, as well as melanoma invasion, pancreatic lipid reprogramming, wound scarring, and tissue inflammation. Therapeutic studies generally favor dampening or redirecting PIEZO1 activity through knockdown, cholesterol manipulation, biomaterials, or natural products rather than treating it as a universal activator. How state-specific gating and cell context determine whether PIEZO1 promotes repair or pathology remains open.
In cancer models, activating PIEZO1 can be therapeutic rather than pathogenic
The pancreatic-cancer model treated with MβCD@HA-Ad nanoparticles and the tumor-therapy model treated with Janus ACC@SiO₂&mPDA-Arg-HA nanomotors both use PIEZO1 activation to increase intracellular Ca²⁺ and suppress tumors, rather than treating PIEZO1-driven Ca²⁺ influx as a process to inhibit 42532144Jul 42411126Jul. This reverses the predominant assumption in the set that mechanically induced PIEZO1 activation promotes disease by driving migration, barrier disruption, scarring, vascular instability, or systemic inflammation. The resulting change is therapeutic: PIEZO1 stimulation and consequent Ca²⁺ overload become an antitumor strategy in these models.
The PIEZO1 wedge domain acts as a state-dependent inactivation particle
The purified-channel and cellular electrophysiology model examined by site-directed mutagenesis and MINFLUX nanoscopy identifies the PIEZO1 intracellular wedge domain as a structural regulator of channel inactivation 42649207Aug. Whereas the other papers use PIEZO1 primarily as a mechanosensor whose activation alters disease, migration, repair, or tumor-cell behavior, this study assigns it a distinct molecular role: wedge docking stabilizes the flat inactivated state, and wedge dissociation permits recovery from inactivation. This redirects the question from whether PIEZO1 should be activated or inhibited to how its conformational state terminates and resumes channel activity.
