Insulin-like growth factor 1
Insulin-like growth factor 1 (IGF-1) is a circulating growth factor and a clinical biomarker of the growth hormone (GH)–IGF-1 axis.
Insulin-like growth factor 1 (IGF-1) is a circulating growth factor and a clinical biomarker of the growth hormone (GH)–IGF-1 axis. Serum IGF-1 concentrations are used in studies of growth, nutritional and metabolic status, and recovery from illness. Recent work has examined IGF-1 in children with persistent diarrhea, in omnivorous, vegetarian, and vegan children, and in people with anorexia nervosa, including changes associated with weight restoration and clinical treatment. Its relationship with cognitive impairment has also been investigated in older patients with cerebral small-vessel disease.
IGF-1 acts through the IGF-1 receptor (IGF-1R) and has been studied in vascular, neural, and tissue-repair contexts. Experimental studies implicate endothelial IGF-1/IGF-1R signaling in restoration of the blood–brain barrier and microvascular architecture in aged mice, while related work examines IGF-1-dependent regulation of pericytes and the blood–labyrinth barrier in age-related hearing loss through interaction with TGF-β/Smad2 signaling. Engineered IGF-1 delivery, including IGF-1-secreting cells and controlled-release biomaterials, is being investigated for diabetic wounds, liver bleeding, and intervertebral-disc repair. These applications remain primarily experimental and context-specific.

Growth Factor Physiology and Repair
IGF-1 links nutritional growth status with vascular integrity, neural health, and tissue repair across the literature. Human studies associate IGF-1-axis variation with diarrhea-related growth impairment, dietary pattern, anorexia nervosa, and cognitive risk. Preclinical work implicates IGF-1/IGF-1R signaling in blood-brain and blood-labyrinth barriers, pericyte function, and endothelial recovery. Engineered IGF-1 delivery extends this work toward liver, diabetic-wound, and disc repair, but clinical applications remain context-specific.
